Abstract
One striking aspect of immunodeficiency viruses istheir ability to replicate and maintain persistent infectionin the face of the host immune response and to eventuallyovercome this response (Desrosiers 1999). The progressive loss of CD4+ T lymphocytes, the hallmark of AIDSinfection, reflects the host's inability to regenerate thesecells as immunodeficiency viruses disrupt the normalfunction of the lymphoid organs. The ensuing havocwrought on the immune system results, perhaps directly,from viral gene expression in the infected cells. Unlikesimple retroviruses, the genomes of human and simianimmunodeficiency viruses (HIV and SIV) encode severalauxiliary proteins that modulate the normal function ofthe infected cells to maximize viral replication in the infected host, and these factors are important determinantsof the clinical outcome of immunodeficiency virus infection (Cullen 1998; Desrosiers et al. 1998; Emerman andMalim 1998). Some of these proteins, such as Nef andVpu, interact with the protein sorting and general signaltransduction machineries in T cells and macrophages (Peter 1998). Other auxiliary proteins, such as Vpr and Vpx,interact with the cell cycle and nuclear transport machineries.
