Abstract

Renewed interest and impetus has been given to the study of the structure of rhodopsin by the recent demonstration that it is a member of a new but probably fairly widespread class of membrane-bound protein receptors. It is likely that all the members of the club will share major structural and conformational features. To understand their individual functional characteristics, a high-resolution structure is very urgently required and can be provided only by crystallographic methods. Many of the important functional properties of these proteins, however, can be very usefully investigated by techniques in protein engineering. The model we describe here has arisen mainly through the application of protein chemistry. Although crude and, at least in detail, probably inaccurate, it nevertheless raises some interesting possibilities and questions which can be addressed by protein engineering. At the same time, it can provide a framework to test the consequences of site-directed mutations. In addition, a whole new area of pharmacological relevance is opened up since we can now begin to marry the structures of agonists and antagonists with their protein targets. The information which results from these types of study can then be used to refine or correct the model. There can hardly be any better example of the inter-disciplinary nature of modern science and of the importance of molecular recognition.
