Abstract
The action of solutions of purified, crystalline trypsin at various pH's on unfixed and fixed salivary-gland chromosomes of Drosophila melanogaster is described and illustrated. Chromosomes of glands transferred directly from physiological salt solution to buffered solutions of trypsin showed no evidence of enzymatic degradation during the first few hours of treatment. Chromosomes of fixed cells were readily hydrolyzed by trypsin dissolved in either water or buffer. Structural deformation did not occur during hydrolysis in aqueous trypsin, but was induced by subsequent treatment with phosphate buffer, or during the course of hydrolysis in buffered solutions. The amount of deformation was found to depend on the pH and duration of the treatment. Structural deformation is attributed to the swelling of partially degraded nucleoproteins in the presence of electrolytes, since removal of desoxyribonucleic acid from the chromosomes inhibited swelling. The application of the findings to problems of structural organization is discussed.
